Les propri\u00e9t\u00e9s antifongiques et antioxydantes des compos\u00e9s synth\u00e9tis\u00e9s ont \u00e9galement \u00e9t\u00e9 \u00e9valu\u00e9es et les r\u00e9sultats \u00e9taient prometteurs. Les \u00e9tudes de docking ont montr\u00e9 qu&rsquo;un produit de la s\u00e9rie synth\u00e9tis\u00e9e a une forte affinit\u00e9 pour la prot\u00e9ine N-myristoyltransf\u00e9rase (NMT) et se lie au site actif de la prot\u00e9ine de la m\u00eame mani\u00e8re que les inhibiteurs utilis\u00e9s en m\u00e9decine et pourrait faire partie de nouvelles classes d&rsquo;agents antifongiques cliniquement efficaces. Visant \u00e0 synth\u00e9tiser des produits hautement fonctionnels, la synth\u00e8se d\u2019une nouvelle s\u00e9rie de mol\u00e9cules d\u00e9riv\u00e9es d&rsquo;isonicotinoylpyrazolylcoumarine par la strat\u00e9gie d&rsquo;hybridation mol\u00e9culaire \u00e0 partir des d\u00e9riv\u00e9s de la 3-ac\u00e9toac\u00e9tylcoumarine avec l&rsquo;isoniazide a \u00e9t\u00e9 r\u00e9alis\u00e9e dans un second temps. Les compos\u00e9s obtenus ont \u00e9t\u00e9 \u00e9valu\u00e9s pour leurs activit\u00e9s antifongiques et deux produits ont montr\u00e9 des activit\u00e9s inhibitrices potentiellement \u00e9lev\u00e9es. Les r\u00e9sultats de l&rsquo;activit\u00e9 antioxydante \u00e9taient \u00e9galement satisfaisants. Les \u00e9tudes relations structure-activit\u00e9 ont d\u00e9montr\u00e9 que la pr\u00e9sence de groupes nitro et hydroxyle dans le cycle aromatique des cycles coumarine et pyrazoline conf\u00e9rait une activit\u00e9 antioxydante et antifongique accrue. En outre, des \u00e9tudes de docking mol\u00e9culaire ont \u00e9t\u00e9 r\u00e9alis\u00e9es pour le compos\u00e9 pr\u00e9sentant une bonne activit\u00e9 antifongique afin d&rsquo;\u00e9valuer son potentiel en tant qu&rsquo;inhibiteur de la prot\u00e9ine fongique (Phytase A) d&rsquo;<em>Aspergillus niger <\/em>et les r\u00e9sultats sugg\u00e8rent que notre isonicotinoylpyrazolylcoumarine peut agir comme un agent antifongique prometteur.<\/p>\n<p style=\"text-align: justify\"><strong>Mots cl\u00e9s\u00a0: <\/strong>H\u00e9t\u00e9rocycle, Coumarine, Dod\u00e9cylbenz\u00e8nesulfonate de sodium (DBSS), Benzylpyrazolylcoumarine, R\u00e9action multicomposants, Hydrazide d&rsquo;acide isonicotinique, Pyrazoline, Activit\u00e9 antioxydante, Activit\u00e9 antifongique, Docking mol\u00e9culaire, prot\u00e9ine N-myristoyltransf\u00e9rase (NMT), prot\u00e9ine fongique (Phytase A) d&rsquo;Aspergillus.<\/p>\n<p>[\/et_pb_accordion_item][et_pb_accordion_item title=\u00a0\u00bbR\u00e9sum\u00e9 de la th\u00e8se en Anglais\u00a0\u00bb open=\u00a0\u00bboff\u00a0\u00bb _builder_version=\u00a0\u00bb4.19.5&Prime; _module_preset=\u00a0\u00bbdefault\u00a0\u00bb body_text_color=\u00a0\u00bb#000000&Prime; global_colors_info=\u00a0\u00bb{}\u00a0\u00bb]<\/p>\n<p><strong>Abstract<\/strong><\/p>\n<p style=\"text-align: justify\">This thesis focuses in its first part, on the synthesis of a series of benzylpyrazolylcoumarin derivatives, involving the multicomponent reaction of 4-hydroxycoumarin, ethyl acetoacetate, hydrazine hydrate and benzaldehyde derivatives, using sodium dodecylbenzenesulfonate (SDBS) as catalyst. The antifungal and antioxidant properties of the synthesized compounds were also evaluated and the results were promising. Docking studies showed that one product of the synthesized series has a high affinity for the protein N-myristoyltransferase (NMT) and binds to the active site of the protein in the same way as inhibitors used in medicine and could be part of new classes of clinically effective antifungal agents. Aiming to synthesize highly functional products, the synthesis of a new series of isonicotinoylpyrazolylcoumarin-derived molecules by the molecular hybridization strategy from 3-acetoacetylcoumarin derivatives with isoniazid was performed in a second step. The obtained compounds were evaluated for their antifungal activities and two products showed potentially high inhibitory activities. The results of the antioxidant activity were also satisfactory. Structure-activity relationship studies demonstrated that the presence of nitro and hydroxyl groups in the aromatic ring of the coumarin and pyrazoline rings conferred enhanced antioxidant and antifungal activity.
